ラパマイシンの投与で成体マウスの自閉症様症状が回復

UCLAヘルスによる研究で、ラパマイシンの単回投与が成体マウスの自閉症様の脳の変化や行動を約2時間以内に改善させることが判明した。ただし、その効果は一時的なものであり、研究はマウスのみを対象に行われた。

ネイチャー・コミュニケーションズに掲載された本研究では、マウスの妊娠中に軽度の炎症が生じると、その子孫に脳の過活動、感覚過敏、常同行動、発作リスクの上昇といった長期的な問題が引き起こされることが示された。研究チームは、免疫抑制剤であるラパマイシンを成体となった子孫に単回投与したところ、神経活動が迅速に正常化し、発作の脆弱性が低下し、脳のネットワーク間のコミュニケーションが改善した。

UCLA知的・発達障害研究センターの所長であり、本研究の上席著者であるハーレー・コーンブラム医師は、機能が急速に正常化したことは、成人の脳がこれまでの想定よりも順応性が高い可能性を示唆していると述べた。筆頭著者であるジャネル・ル・ベル医師は、自閉症に関連する一部の特性は、物理的な構造ではなく機能的な回路をターゲットにすることで対処できる可能性があると指摘した。

共同上席著者のニール・ハリス医師は、ラパマイシンの効果は持続せず、繰り返しの使用は毒性を示す可能性があるため、人間への治療法としては適さないと警告した。しかし、今回の知見は、将来的な治療法としてmTOR経路の活性や神経バランスといった潜在的な標的の存在を示唆している。
