ラット研究、ルームメイトの遺伝子が腸内細菌に影響することを示す

数千匹のラットに関する新たな研究では、社会的パートナーの遺伝子が共有微生物を通じて個体の腸内マイクロバイオームを形成し得ることが示唆された。研究者らは、これらの社会的効果を考慮すると遺伝的影響が強まることが分かった。この知見は、微生物交換を介した遺伝子が健康に間接的に影響を与える方法を強調している。

12月18日にNature Communicationsに掲載されたこの研究では、米国異なる施設で飼育された4つのコホートから4,000匹以上の遺伝的にユニークなラットを調査した。これらの制御された条件により、科学者たちは消化と健康を助ける兆単位の微小生物からなる腸内マイクロバイオームに対する遺伝的影響を分離できた。

バルセロナのCentre for Genomic Regulationとカリフォルニア大学サンディエゴ校のチームは、環境を超えて特定の細菌と一貫して関連する3つの遺伝子領域を特定した。腸粘液を糖で修飾する遺伝子St6galnac1は、Paraprevotella細菌のレベルが高いことと相関した。腸の保護層を形成するムチン遺伝子はFirmicutes細菌と関連し、抗菌分子を産生するPip遺伝子はMuribaculaceae科のメンバーとつながっていた。

微生物共有による間接的な遺伝的効果——1匹のラットの遺伝子がケージメイトに影響を与える——をモデル化することで、この研究はこれらの関連性について社会的要因が遺伝的影響を4〜8倍に増幅することを明らかにした。「これは魔法ではなく、社会的接触を通じて他者に遺伝的影響が波及する結果です。遺伝子は腸内マイクロバイオームを形成し、私たちの遺伝子だけが重要ではないことが分かりました」と、主任著者のAmelie Baud博士は述べた。

結果は、DNA転移なしに他者の生物学を間接的に変化させる遺伝子を示唆している。ヒトでは、同様のメカニズムが免疫機能や代謝などの健康問題における遺伝的役割を過小評価する可能性がある。ラットのSt6galnac1はヒトのST6GAL1と関連し、以前にParaprevotellaやCOVID-19感受性、IgA腎症などの状態と関連付けられていた。Baud博士は「おそらく氷山の一角しか明らかにしていない」と指摘した。
