Läkemedelskombination kyler ner kroppen för att begränsa stroke-skador

Två befintliga läkemedel kylde ner kroppstemperaturen hos djur efter stroke och minskade hjärnskador. En liten studie på människor visade begränsad effekt på grund av långsam administrering. Forskare planerar nu en uppföljande studie med snabbare infusioner.

Forskare testade prometazin och klorpromazin på möss och rhesusapor efter att ha inducerat stroke. Läkemedlen sänkte kroppstemperaturen, minskade cellernas glukosmetabolism och reducerade hjärnskador. De behandlade aporna uppvisade även förbättrad lemfunktion.

I en klinisk studie med 32 personer sänkte samma läkemedel temperaturen med endast 0,3°C och minskade inte stroke-skadorna. Forskarlaget tillskrev detta de långsamma 12-timmarsinfusionerna som inte lyckades uppnå effektiva läkemedelsnivåer tillräckligt snabbt.

Läkemedlen, som redan är godkända för hösnuva och psykos, verkar på det centrala nervsystemet utan att orsaka frossa. En ny studie kommer att testa en timmes infusioner för att avgöra om snabbare nedkylning förbättrar resultaten.
