神経細胞の骨格がアルツハイマー病治療の新たな標的となる可能性

ペンシルベニア州立大学の研究チームが、神経細胞内の格子状構造が細胞への取り込みを制御するゲートキーパーの役割を果たしていることを突き止めた。この構造が弱まると、神経細胞はアルツハイマー病に関連する有害なタンパク質をより多く取り込んでしまう。この構造を安定化させることが、新たな治療戦略につながる可能性がある。

膜結合型の周期的な骨格であるMPS(membrane-associated periodic skeleton)は、神経細胞の表面直下に存在する。これは、細胞が周囲から物質を取り込むプロセスであるエンドサイトーシスを制御している。

実験の結果、MPSを破壊すると神経細胞が物質を取り込む速度が速まることが明らかになった。これには、アルツハイマー病に関連する毒性断片に分解されるアミロイド前駆体タンパク質も含まれる。

本研究の責任著者であるRuobo Zhou氏は「この膜骨格が神経細胞の栄養取り込みプロセスを積極的に制御していることを発見した」と述べた。筆頭著者のJinyu Fei氏は、今回の知見がこの構造を維持することを目的とした治療法への扉を開く可能性があると付け加えた。

米国国立衛生研究所(NIH)の助成を受けた本研究は、『Science Advances』誌に掲載された。研究チームは、実験室で培養した神経細胞に対し超解像イメージング技術を用い、その影響を追跡した。
