Medicamentos orais de GLP-1 reduzem sinais de recompensa alimentar em camundongos

Um novo estudo mostra que agonistas orais do receptor de GLP-1 podem controlar a alimentação movida pelo prazer em camundongos, agindo em um circuito de recompensa cerebral. A pesquisa, apoiada pelos Institutos Nacionais de Saúde (NIH), examinou medicamentos incluindo orforglipron e danuglipron.

Pesquisadores da Universidade da Virgínia descobriram que esses medicamentos de moléculas pequenas ativaram a amígdala central, uma região envolvida no desejo e na recompensa. Essa ativação reduziu a liberação de dopamina durante a alimentação hedônica, ou seja, comer por prazer em vez de por necessidades energéticas.

As descobertas vêm de experimentos com camundongos editados geneticamente para possuírem receptores de GLP-1 semelhantes aos humanos. Os medicamentos também influenciaram áreas ligadas à regulação do apetite, mas o efeito no circuito de recompensa mais profundo não era reconhecido anteriormente.

"Sabíamos que os medicamentos de GLP-1 suprimem o comportamento alimentar movido pela demanda de energia. Agora, parece que os GLP-1 orais de moléculas pequenas também reduzem a alimentação por prazer ao envolver um circuito de recompensa cerebral", afirmou o coautor correspondente Ali Guler.

Lorenzo Leggio, do Instituto Nacional sobre Abuso de Drogas, destacou a importância de compreender esses mecanismos à medida que o acesso à medicação aumenta. O estudo foi publicado na revista Nature e não se trata de um ensaio clínico.
