全ゲノム重複(WGD:細胞内のDNA量が通常の2倍になる現象)は、細胞機能不全や遺伝的不安定性といった有害な結果をもたらすものとして、長年研究されてきた。北海道大学による新しい研究は、WGDを引き起こす「メカニズム」がその後の細胞の運命を決定づける主要な要因であると指摘している。

研究チームは、WGDに至る一般的な2つの経路を比較した。

  • 細胞質分裂不全:細胞が体細胞分裂のほとんどの過程を終えたものの、最終的に2つの細胞へ物理的に分離することに失敗する。
  • 有糸分裂の脱落:染色体が適切に分離される前に、細胞が有糸分裂の過程から早期に離脱してしまう。

ライブセルイメージングと染色体標識の手法を用いた分析の結果、細胞質分裂不全によって生じた細胞は比較的安定しており生存率が高い一方、有糸分裂の脱落によって生じた細胞は、染色体の不均等な分配がより頻繁に見られ、生存率も低いことが明らかになった。

また、実験的に有糸分裂の脱落を起こす細胞の染色体分離を改善したところ、細胞の生存率が大幅に向上した。このことから、染色体の構成と分離の正確さが、細胞の運命を分ける鍵であることが示唆された。

著者らは、この知見ががん研究に示唆を与える可能性があると述べている。がん細胞では全ゲノム重複が一般的に観察されるほか、一部の治療法によって意図せずWGDが誘発されることもあるためである。今回の結果は、WGD後に生き残る可能性が高い細胞が、がんの増殖や再発を促進している可能性があることを示している。
